The first two lines are in reference to the Hughes Bros. classic “Menace 2 Society.” Caine’s dad played by Samuel Jackson (Tat) shoots a guy at a poker game for telling him he’s going to pay him “when he feels like it.” Before shooting him, he says to Tat “Who do you think you are, Ron O'Neal?” So when Tat pulls out the joint on him, the guy asks him “Am I supposed to be scared or something?”

Nicki may be comparing herself to NY Giant’s quarterback Eli Manning, because both are incredibly talented (multiple Grammys/Super Bowl wins respectively), but are constantly outshined by their older brothers. Nicki is often overshadowed by Young Money labelmates Drake & Lil Wayne, while Eli is overshadowed by his actual brother, Peyton Manning.

Possibly also a short-joke aimed at Lil' Kim. Eli Manning plays for The New York Giants – emphasis on the term Giant here. Nicki is especially calling herself a “Giant” in height [when compared to Kim] and in the rap game, while compared to how Lil' Kim is.
